What is the ruling regarding a wife who defied her husband's orders, then left the house after her husband intended to discipline her for her mistake, knowing that her mistake was based on a misunderstanding of a conversation with her mother-in-law?

The penitent wife is obligated to return to the obedience of her husband and not to rush into accepting everything she hears. If the wife's departure is for a legitimate Sharia (definition) reason, it is not considered an act of defiance (nushooz), such as her fearing for herself from an unlawfully authorized punishment. The husband should address his wife's mistakes with wisdom, bearing in mind that women were created from a rib, and that the marital relationship may experience some annoyances. The Prophet, peace and blessings be upon him, said: "No believing man should hate a believing woman; if he dislikes one of her characteristics, he will be pleased with another."
